The linac coherent light source single particle imaging road map
نویسندگان
چکیده
Intense femtosecond x-ray pulses from free-electron laser sources allow the imaging of individual particles in a single shot. Early experiments at the Linac Coherent Light Source (LCLS) have led to rapid progress in the field and, so far, coherent diffractive images have been recorded from biological specimens, aerosols, and quantum systems with a few-tens-of-nanometers resolution. In March 2014, LCLS held a workshop to discuss the scientific and technical challenges for reaching the ultimate goal of atomic resolution with single-shot coherent diffractive imaging. This paper summarizes the workshop findings and presents the roadmap toward reaching atomic resolution, 3D imaging at free-electron laser sources.

Coherent soft X-ray diffraction imaging of coliphage PR772 at the Linac coherent light source
Single-particle diffraction from X-ray Free Electron Lasers offers the potential for molecular structure determination without the need for crystallization. In an effort to further develop the technique, we present a dataset of coherent soft X-ray diffraction images of Coliphage PR772 virus, collected at the Atomic Molecular Optics (AMO) beamline with pnCCD detectors in the LAMP instrument at t...
متن کاملCoherent diffraction of single Rice Dwarf virus particles using hard X-rays at the Linac Coherent Light Source
Single particle diffractive imaging data from Rice Dwarf Virus (RDV) were recorded using the Coherent X-ray Imaging (CXI) instrument at the Linac Coherent Light Source (LCLS). RDV was chosen as it is a well-characterized model system, useful for proof-of-principle experiments, system optimization and algorithm development. RDV, an icosahedral virus of about 70 nm in diameter, was aerosolized an...
متن کاملMerging single-shot XFEL diffraction data from inorganic nanoparticles: a new approach to size and orientation determination
X-ray free-electron lasers (XFELs) provide new opportunities for structure determination of biomolecules, viruses and nanomaterials. With unprecedented peak brilliance and ultra-short pulse duration, XFELs can tolerate higher X-ray doses by exploiting the femtosecond-scale exposure time, and can thus go beyond the resolution limits achieved with conventional X-ray diffraction imaging techniques...
متن کاملDragonfly: an implementation of the expand–maximize–compress algorithm for single-particle imaging1
Single-particle imaging (SPI) with X-ray free-electron lasers has the potential to change fundamentally how biomacromolecules are imaged. The structure would be derived from millions of diffraction patterns, each from a different copy of the macromolecule before it is torn apart by radiation damage. The challenges posed by the resultant data stream are staggering: millions of incomplete, noisy ...
متن کاملLinac Design for the LCLS Project at SLAC
The Linac Coherent Light Source (LCLS) at SLAC is being designed to produce intense, coherent 0.15-nm x-rays. These x-rays will be produced by a single pass of a 15-GeV bunched electron beam through a long undulator. Nominally, the bunches have a charge of 1 nC, normalized transverse emittances of less than 1.5π mm-mr and an rms bunch length of 20 μm. The electron beam will be produced using th...
